Emma06 Posted March 14, 2021 Share Posted March 14, 2021 Salut ! Que la chromatographie soit d'affinité/ d'exclusion/ échangeuse d'ion ..etc, est ce que elle se fait en condition dénaturante ? Parce que du coup par exemple pour la chromatographie d'exclusion si on a une protéine trimérique (avec chaque monomère qui fait une taille différente) on aura 3 pics d'absorbance ou bien 1 seul ? Je confirme ce qu'à dit @E182, si c'est pas précisé, on est en conditions non-dénaturante. En général, on se sert d'une chromato pour purifier notre protéine et on veut garder son activité. Donc habituellement, on évite de la dénaturer pour qu'elle puisse continuer à fonctionner. Donc pour une protéine trimérique, tu n'auras qu'un seul pic sur ta chromato d'exclusion.
