Le récepteur NMDA au glutamate est un peu particulier, son activation se fait en 2 temps et nécessite une dépolarisation :
- tout d'abord il y a fixation du glutamate (ou tout autre agoniste) sur sa sous-unité, ce qui permet le départ de l'ion Mg2+ : cet ion, situé dans le canal, agit comme une sécurité, il va empêcher tout passage possible d'ions en dehors de la dépolarisation
