maestro Posted May 5, 2019 Share Posted May 5, 2019 Bonsoir par rapport à l'item D, je ne comprends pas comment on peut conclure que la protéine FLIP est dépourvu d'activité sur le substrat, car certe elle ne libère par l'AMC ( on le voit car pas d'activité fluorescente dans la seconde colonne), mais elle pourrait avoir une autre activité sur ce substrat, jsp genre par exemple elle pourrait couper après la Valine, mais elle ne libere donc pas l'AMC, bref je ne vois pas comment on peut conclure.. Une caspase est un Cysteinyl Aspartate Proteinase ce qui veut dire qu’elle peut couper après un Aspartate si le site catalytique possède une Cysteine. Du coup si la protéine FLIP possédait une activité caspase alors elle couperai forcément le substrat fluorogénique après l’aspartate et libererai l’AMC. Tu suis ? Voilà bon courage pour la suite !!
